c-Raf - Wikipedia
C-Raf is a member of the Raf kinase family of serine/threonine-specific protein kinases, from the TKL (Tyrosine-kinase-like) group of kinases. The first Raf gene, v-Raf was found in 1983. It was isolated from the murine retrovirus bearing the number 3611.
Raf Kinases: Function, Regulation and Role in Human Cancer
The human C-Raf (Raf-1 or c-Raf-1), B-Raf and A-Raf are aligned with the Drosophila Raf (D-Raf) and C.elegans Raf (lin45). Indicated are the positions of conserved regions (CR) 1, 2 and 3, the Ras binding domain (RBD), the zinc finger/cysteine rich domain (CRD) and the kinase domain.
The role of CRAF in cancer progression: from molecular ... - Nature
2024年1月9日 · Here, we summarize the main known regulatory mechanisms and gene alterations that contribute to CRAF activity, highlighting the different oncogenic roles of CRAF, and categorize RAF1 (CRAF)...
C-Raf - an overview | ScienceDirect Topics
C-Raf is a serine/threonine kinase that is part of the Raf family, which is activated by ras-GTP and/or PKC-α. It plays a crucial role in transducing signals along the Ras-Raf-MEK-ERK pathway, making it a potential target for anticancer therapy, especially in tumors with activating mutations in …
Mutations of C-RAF Are Rare in Human Cancer because C-RAF …
2005年11月1日 · Here we show that presumptive mutants of the closely related kinase, C-RAF, were detected in only 4 of 545 (0.7%) cancer cell lines. The activity of two of the mutated proteins is not significantly different from that of wild-type C-RAF and these variants may represent rare human polymorphisms.
Raf kinases in cancer–roles and therapeutic opportunities
2011年5月16日 · Raf are conserved, ubiquitous serine/protein kinases discovered as the cellular elements hijacked by transforming retroviruses. The three mammalian RAF proteins (A, B and CRAF) can be...
C-Raf is Associated with Disease Progression and Cell Proliferation …
2009年9月9日 · We studied effects of C-Raf knock-down in vitro and assessed expression of C-Raf in a large cohort of melanomas and nevi. Using specific siRNAs, we knocked down C-Raf expression, and determined the effect on viability, MEK/ERK signaling and apoptosis in seven melanoma cell lines.
RAF-1 (C-RAF) | SpringerLink
Raf-1, also known as C-Raf-1 or C-Raf, was identified about 30 years ago as the oncogene (v-raf) in the murine sarcoma virus 3611 (3611-MSV) and, in parallel, in the naturally occurring avian retrovirus Mill Hill 2 (MH2).
Cardiac-specific disruption of the c-raf-1 gene induces cardiac ...
2004年10月1日 · In this study, we generated cardiac-specific Raf-1 knockout (Raf CKO) mice to clarify the role of Raf-1 in the heart. These mice showed left ventricular (LV) dysfunction and heart dilatation with an increased number of apoptotic cells. These phenotypes were rescued by inactivation of ASK1.
Activation of c-Raf-1 by Ras and Src through different …
The c-Raf-1 protein kinase plays a critical role in intracellular signaling downstream from many tyrosine kinase and G-protein-linked receptors. c-Raf-1 binds to the proto-oncogene Ras in a GTP-dependent manner, but the exact mechanism of activation of c-Raf-1 by Ras is still unclear.
