Paired box gene 8, also known as PAX8, is a protein which in humans is encoded by the PAX8 gene. [5] This gene is a member of the paired box (PAX) family of transcription factors. Members of this gene family typically encode proteins which contain a paired box domain, an octapeptide, and a paired-type homeodomain.

PAX8 has been demonstrated to be crucial in determining cell fate during the development of the thyroid, kidney, brain, eyes and Müllerian system and regulates expression of the Wilms’ tumor suppressor gene (WT1).

Following birth, the PAX8 protein regulates several genes involved in the production of thyroid hormones. At least 15 mutations in this gene cause congenital hypothyroidism, a condition characterized by abnormally low levels of thyroid hormones starting from birth.
