2016年3月24日 · We recently found that focal adhesion kinase 1 (FAK1) is upregulated and phosphorylated (active) in IBC. In this study, we investigated the effect of CEP-37440, a dual inhibitor of FAK1 and anaplastic lymphoma kinase (ALK), using human IBC cell lines and preclinical models of IBC.

Since we identified FAK1 as a topological and functional hub kinase of the 36 kinases, we sought to determine how FAK1 IDRs can be regulated, which, in turn, can regulate function of FAK1. FAK1 protein structure consists of three domains: FERM domain, KD, and FAT domain.

Pharmacological inhibition with FAK1 inhibitor FAK14 attenuated LPS-induced endothelial inflammatory activation and leukocyte adhesion partly via blockade of NF-κB activity. Administration of FAK14 after EC exposure to LPS also resulted in inhibition of inflammatory molecule expression.
