Diclofenac is a non-steroidal anti-inflammatory drug (NSAID) used for the relief of pain, swelling, and joint stiffness caused by conditions such as arthritis. It comes in different formulations, including tablets, gels, and topical solutions.
Diclofenac was the product of rational drug design based on the structures of [phenylbutazone], [mefenamic acid], and [indomethacin]. The addition of two chlorine groups in the ortho position of the phenyl ring locks the ring in maximal torsion which appears to be related to increased potency.

Diclofenac sodium is a non-selective reversible and competitive inhibitor of cyclooxygenase (COX), subsequently blocking the conversion of arachidonic acid into prostaglandin precursors. This leads to an inhibition of the formation of prostaglandins that are involved in pain, inflammation and fever.
