Carcinoembryonic antigen-related cell adhesion molecule 8 (CEACAM8) also known as CD66b (Cluster of Differentiation 66b), is a member of the carcinoembryonic antigen (CEA) gene family. Its main function is cell adhesion, cell migration, and pathogen binding. [3]

The CD66b + monocytes displayed high phagocytic activity, matrix adhesion and migration, and provided costimulation for T cell proliferation and IFN-γ secretion; thus, they did not suppress T cell responses. Irrespective of clinical stage, they were identified in various cancers.

CD66b, a granulocyte activation marker typically found on neutrophils and eosinophils, may have predictive value in patients treated with immunotherapy.

Directly isolated neutrophils express very high levels of CD66b. It is also expressed by eosinophils. See supplemental figure S4H in this recent Immunity paper

CD66b is thought to play a role in the regulation of the adhesive activity of CD11/CD18 via signal transduction in neutrophils. It is strongly expressed by myelocytes, metamyelocytes, neutrophils and polymorphonuclear neutrophils. CD66b is weakly expressed by promyelocytes.

Neutrophils express several heavily glycosylated carcinoembryonic antigen (CEA)-related glycoproteins (CD66 antigens) which have been implicated in adhesion to E-selectin and as receptors for the lectins galectin 3 and bacterial type-1 fimbriae.

CD66b is highly expressed on the surface of peripheral blood eosinophils . It is expressed by promyelocytes and early myelocytes, reaches maximal expression at the late myelocyte and metamyelocyte stages and then decreases at the band and segmented neutrophil stages [ 11 ].
