Necdin is a protein that in humans is encoded by the NDN gene. [5][6] This intronless gene is located in the Prader-Willi syndrome (PWS) deletion region. It is an imprinted gene and is expressed exclusively from the paternal allele. Studies in mice suggest that the protein encoded by this gene may suppress growth in postmitotic neurons. [6]

Necdin was originally found in 1991 as a hypothetical protein encoded by a neural differentiation-specific gene transcript in murine embryonal carcinoma cells. Virtually all postmitotic neurons and their precursor cells express the necdin gene (Ndn) during neuronal development.

Necdin is a protein that is functionally similar to Rb, but structurally different. It is a smaller protein and does not undergo posttranslational modifications. Necdin plays a role in suppressing cell growth and binds to various proteins, including the transactivation domains of E2F1 and p53.
