2017年4月28日 · In this article, we review the current understanding of protein transport across the ER membrane as central function of Sec proteins and further focus on recent studies that gave first insights...
A common mechanism of Sec61 translocon inhibition by small …
2023年5月11日 · Here we present near-atomic-resolution structures of human Sec61 inhibited by a comprehensive panel of structurally distinct small molecules—cotransin, decatransin, apratoxin, ipomoeassin,...
An Update on Sec61 Channel Functions, Mechanisms, and Related …
2017年11月1日 · Recently, cryoelectron tomography of translocons in native ER membrane vesicles, derived from human cell lines or patient fibroblasts, and even intact cells has given unprecedented insights into the architecture and dynamics of …
An Update on Sec61 Channel Functions, Mechanisms, and Related …
These structural data are discussed in light of different Sec61 channel activities including ribosome receptor function, membrane insertion, and translocation of newly synthesized polypeptides as well as the putative physiological roles of the Sec61 channel as a passive ER calcium leak channel.
Sec61 complex/translocon: The role of an atypical ER Ca
The Sec61 complex appears to be a good candidate to explain the mechanism of Ca 2+-store depletion associated with ER stress and UPR. The role of the Sec61 complex in cell death and ER stress has been investigated in the human cancerous cell line LNCaP (Hammadi et al., 2013).
Sec61 - an overview | ScienceDirect Topics
The Sec61 complex provides a high-affinity docking site for the ribosome–nascent chain complex. Additional factors thought to facilitate binding of the signal sequence to the Sec61 complex include the protein TRAM (translocating chain-associating membrane protein) and the protein complex TRAP (translocon-associated protein).
Mechanism of Protein Translocation by the Sec61 Translocon …
2023年1月3日 · The universally conserved protein-conducting channel Sec61 complex mediates core steps in these processes by translocating hydrophilic polypeptide segments of client proteins across the ER membrane and integrating hydrophobic transmembrane segments into …

2017年4月28日 · In this article, we review the current understanding of protein transport across the ER membrane as central function of Sec proteins and further focus on recent studies that gave first insights into the functional role and therapeutic relevance of Sec61, Sec62 and Sec63 in human diseases.
Structure of the Sec61 channel opened by a signal sequence
2016年1月1日 · Here, we use electron cryo-microscopy to determine the structure of an active Sec61 channel that has been opened by a signal sequence. The signal supplants helix 2 of Sec61α, triggering a rotation that opens the central pore both axially across the membrane and laterally toward the lipid bilayer.
